Aims: This study aimed to investigate the combined effect of eight weeks of HIIT and resveratrol supplementation on DNA damage (8-hydroxydeoxyguanosine; 8-OHdG) and lipid peroxidation (malondialdehyde; MDA) in type 2 diabetic rats.
Materials and Methods: Forty male Wistar rats were randomly divided into four groups: diabetic control, diabetic+HIIT, diabetic+resveratrol (10 mg/kg/day), and diabetic+HIIT+resveratrol. Type 2 diabetes was induced by a high-fat diet combined with streptozotocin injection (35 mg/kg). The HIIT protocol consisted of 6-12 intervals of 2 minutes at 85-90% of maximum speed with 3-minute active recovery, performed five days per week for eight weeks. Serum levels of 8-OHdG and MDA were measured using ELISA and spectrophotometric methods, respectively.
Results: Both HIIT and resveratrol significantly reduced 8-OHdG (P<0.001 and P<0.005, respectively) and MDA (P<0.005 and P<0.001, respectively) compared to the diabetic control group. The combined intervention demonstrated the most substantial reductions in both markers, with significant differences compared to HIIT alone (8-OHdG: P<0.005; MDA: P<0.005) and resveratrol alone (8-OHdG: P<0.001; MDA: P<0.005). No significant differences were observed between HIIT and resveratrol groups for either marker (P=0.487 and P=0.491, respectively).
Conclusion: Both HIIT and resveratrol independently reduce DNA damage and lipid peroxidation in diabetic rats. The combination of these interventions exhibits a synergistic effect, resulting in superior protection against oxidative damage compared to either intervention alone, suggesting a promising non-pharmacological strategy for managing diabetic complications.
